SARS-CoV-2 is a new type of corona virus from the Betacoronavirus genus and Coronaviridae family which causes respiratory disease called COVID-19. SARS-CoV2 is an enveloped virus and has single stranded RNA as its genetic material. The genome structure of this virus consists of two types of protein, first, the gene that encodes non-structural protein such as ORF1a/OR1b, and second, the gene that encodes structural protein such as spike glycoprotein (S), envelope (E), membrane glycoprotein (M), dan nucleocapsid protein (N). Real time RT-PCR test is the most recommended method for SARS-CoV-2 detection because its high specificity and high accuracy results. The test requires samples collected by swab in oropharynx and nasopharynx for further examination in laboratory in, which later the presence of viral RNA becomes a molecule that is assessed for diagnostic results. In this study, volume optimization was carried out on the Da An Kit used for SARS-CoV-2 detection in real time RTPCR with the objective of saving the use of available kit reagents yet with the optimal and accurate amplification results. There were three samples of SARS-CoV-2 RNA used in this study namely N62, N63 and N79 and three different total volumes were tested from 20 �l, 15 �l and 10 �l. The results of this study showed that these three samples positively contained SARS-CoV-2 with Cq values < 40. Beside that, from these three total volumes that have been tested, it was found a total volume of 20 �l was the optimal volume because produced the lowest Cq values compared to the other two volumes
