Description:
Salah satu gen yang mengatur terjadinya apoptosis pada sel adalah gen BCL-2, yang
ekspresi proteinnya berpotensi sebagai penanda prognosis kanker. Pengukuran tingkat
transkripsi gen BCL-2 dapat dilakukan dengan One-step Real-Time PCR (RT-PCR). Metode
ini dapat menggambarkan proses transkripsi gen BCL-2 yang nantinya akan ditranslasikan
menjadi protein. Optimasi reaksi qPCR perlu dilakukan untuk mendapatkan hasil pengukuran
yang valid. Penelitian kali ini dilakukan untuk mengkarakterisasi primer gen BCL-2 sebagai
bagian optimasi reaksi RT-PCR. Sampel yang digunakan adalah RNA dari orang sehat,
diisolasi dengan reagen tripure (Gene Foci, USA ). Reaksi RT-PCR dilakukan menggunakan
SensiFAST SYBR No-ROX Mix (BioLine, USA) menggunakan primer yang didapatkan dari
hasil penelitian sebelumnya. Hasil penelitian menunjukkan bahwa gen BCL-2 dapat
diamplifikasi dengan nilai Ct = 22, menggunakan RNA dengan konsentrasi 382,04 ng/μl. Perlu
dilakukan optimasi lebih lanjut untuk dapat menghasilkan kondisi RT-PCR yang sensitif dan
spesifik pada gen BCL-2
